Peripheral Blood Film Examination.
The Complete Blood Count (CBC) with Peripheral Blood Film (PBF) test is a comprehensive blood test that evaluates overall health by measuring different blood components, including red blood cells (RBC), white blood cells (WBC), hemoglobin, hematocrit, and platelets. The Peripheral Blood Film (PBF) provides a detailed microscopic examination of blood cells to detect abnormalities in shape, size, or structure, helping diagnose infections, anemia, blood disorders, and certain cancers.

Overview
A Complete Blood Count (CBC) with Peripheral Blood Film (PBF) is a comprehensive blood test that evaluates overall health and detects a wide range of disorders, including anemia, infections, and blood cancers.
- CBC measures different components of blood, such as red blood cells (RBC), white blood cells (WBC), hemoglobin (Hb), hematocrit (Hct), and platelets.
- PBF (Peripheral Blood Smear) provides a detailed microscopic examination of blood cells, helping diagnose abnormalities in size, shape, and structure.
Risk Assessment
This test is useful for detecting:
✅ Anemia (Iron deficiency, megaloblastic anemia, hemolytic anemia, etc.)
✅ Infections and Inflammation (Bacterial, viral, or parasitic infections)
✅ Blood Disorders (Leukemia, lymphoma, myelodysplastic syndromes)
✅ Bone Marrow Disorders (Aplastic anemia, polycythemia, thrombocytopenia)
✅ Clotting Disorders (Thrombocytosis, thrombocytopenia).
Normal Range for CBC
(May vary slightly based on laboratory standards)
- RBC: Male: 4.7-6.1 million/µL, Female: 4.2-5.4 million/µL
- WBC: 4,000-11,000 /µL
- Hemoglobin (Hb): Male: 13.8-17.2 g/dL, Female: 12.1-15.1 g/dL
- Hematocrit (Hct): Male: 40.7-50.3%, Female: 36.1-44.3%
- Platelets: 150,000-450,000 /µL.
Interpretation of Results
| Parameter | Normal Range | Interpretation |
|---|---|---|
| Hemoglobin (Hb) | Male: 13.8-17.2 g/dL, Female: 12.1-15.1 g/dL | Low → Anemia, High → Polycythemia |
| Red Blood Cells (RBC) | Male: 4.7-6.1 million/µL, Female: 4.2-5.4 million/µL | Low → Anemia, High → Polycythemia |
| White Blood Cells (WBC) | 4,000-11,000 /µL | High → Infection, inflammation; Low → Bone marrow suppression |
| Platelets (PLT) | 150,000-450,000 /µL | Low → Bleeding risk; High → Clotting risk |
| Hematocrit (Hct) | Male: 40.7-50.3%, Female: 36.1-44.3% | Low → Anemia, High → Dehydration |
| Mean Corpuscular Volume (MCV) | 80-100 fL | Low → Microcytic anemia, High → Macrocytic anemia |
| Mean Corpuscular Hemoglobin (MCH) | 27-33 pg | Low → Hypochromic anemia, High → Hyperchromic anemia |
PBF Findings & Their Significance:
- Microcytosis & Hypochromia → Iron deficiency anemia
- Macrocytosis → Vitamin B12/Folate deficiency
- Schistocytes → Hemolytic anemia
- Target cells → Liver disease, thalassemia
- Blast cells → Leukemia
- Toxic granulation in neutrophils → Severe bacterial infection.
Sample Type & Collection
Sample Type: Venous blood
Collection Method:
- Blood drawn from a vein (usually arm) into an EDTA tube (purple/lavender cap)
- Peripheral blood smear prepared by spreading a drop of blood on a glass slide
- Stained with Wright-Giemsa stain for microscopic examination.
