How to use Arofrez
Take this medicine in the dose and duration as advised by your doctor. Swallow it as a whole. Do not chew, crush or break it. Arofrez may be taken with or without food, but it is better to take it at a fixed time.
How Arofrez works
Arofrez is an aromatase inhibitor. It works by decreasing the amount of estrogen (natural female hormone) produced in the body. This decrease in estrogen can slow down or stop the growth of some breast cancers that need estrogen to grow. In addition, it is also useful in infertility whereby it increases the release of certain hormones to stimulate the release of eggs from the ovaries.
What if you forget to take Arofrez?
If you miss a dose of Arofrez, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular schedule. Do not double the dose.
Quick Tips
- You have been prescribed Arofrez for the treatment of breast cancer or infertility due to anovulation.
- It can be taken with or without food.
- Do not drive or do anything requiring concentration until you know how Arofrez affects you.
- It may cause increased sweating and hot flushes. Try to keep cool by wearing light and airy clothes.
- Your doctor may monitor your cholesterol level and bone mineral density (BMD) regularly as Arofrez can alter their levels.

Administration

Adult Dose
Oral
Advanced or locally advanced breast cancer ; Adjuvant therapy for postmenopausal women with hormone receptor positive early breast cancer
Adult: 2.5 mg once daily.
Hepatic impairment: Reduce dose by 50% in patients with cirrhosis and severe hepatic impairment; recommended dose: 2.5 mg on alternate days.
Contraindication
Premenopausal women and children; hypersensitivity.
Mode of Action
Letrozole competitively binds to the heme group of aromatase, a cytochrome P450 enzyme which catalyzes conversion of androgen to oestrogen, leading to inhibition of the enzyme and a significant reduction in plasma oestrogen levels.
Precaution
Severe renal impairment; severe hepatic impairment; osteoporosis. Caution when driving or operating machinery.
Lactation: not known if excreted in breast milk (for post-menopausal women)
Side Effect
Hot flushes, arthralgia, nausea, vomiting, fatigue, dizziness, headache, dyspepsia, constipation, diarrhoea, anorexia, alopoecia, increased sweating, rash, peripheral oedema, osteoporosis, musculoskeletal pain, vaginal irritation.
Potentially Fatal: Thromboembolic events.
Interaction
Plasma levels reduced by tamoxifen.
